Principal Scientist –Analytical Development

Synthorx, a Sanofi Company, employs a novel synthetic biology platform for the discovery

and development of transformational protein therapeutics for the treatment of cancer and

autoimmune disorders. We are hiring a Principal Scientist with experience in protein

analytics and in-depth knowledge of characterization to join the CMC team to advance drug

candidates through clinical development and accelerate IND-enabling activities. The

qualified candidate will report to the Senior Director of CMC Analytical and Formulation


Key Responsibilities:

• Design and implement methods for the characterization of PEGylated proteins,

mAbs, antibody fragments and proteomics using state of art analytical technologies

including LC/MS and MS/MS

• Apply extensive scientific knowledge and practical experience to develop separation

methods, e.g. RP, IEX, HILIC, SEC and Protein A affinity chromatography for

quantitative and qualitative analysis

• Apply extensive hands-on experience with various instrument and software

including LC-Qtof mass spectrometry, nanoLC-MS/MS, Orbitrap MS, LC-SQD2, UVVIS

spectrophotometer, etc. to support method development and quick data


• Perform LC-MS/MS method development in conjunction with optimal separation

technologies to identify and quantitate Host Cell Proteins (HCPs) supporting HCP

assay development and downstream process development

• Support characterization of PTMs and relative quantitation of unnatural amino acid

incorporation throughout lead to drug candidate steps

• Develop labelled and label-quantitative peptide assays to support fidelity

(misincorporation) and titer methods throughout the product development cycle

• Participate in comparability studies to establish product attribute profile and

support process characterization and validation

• Collaborate with internal teams and external CMOs or CROs and provide support to

different activities as needed

• Manage priorities, plan experiments, conduct analysis and report results to project


• Prepare research reports and presentations for team meetings, and draft laboratory

standard operation procedures


• Ph.D. in analytical chemistry or related disciplines with a minimum of10 years

relevant industry experience

• Substantial scientific knowledge and hands-on experience in protein structure

elucidation, identification and quantitation of related impurities or PTMs and

degradants using LC-MS/MS

• Practical experience in protein or peptide quantitation by mass spectrometry using

a label or label-free technologies

• Masters software and bioinformatic strategies for protein identification and


• Experience in in-vivo and ex-vivo PK/PD assays, metabolite identification,

automation in high throughput LC/MS based assay is preferred

• Working knowledge of HDX-MS is a plus

• Ability to work with different LC-MS systems and software, performing routine

maintenance and troubleshooting

• Programing using Visual Basic, PE Multiprobe HT or Tecan liquid handler is a plus

• Familiarity with ICH, USP and FDA requirements for method development validation

• Ability to work in a fast-paced and team-orientated environment, and commitment

to a collaborative and professional work environment

• Excellent organization skills and ability to multi-task and manage priority

• Excellent computer skills: proficiency with Microsoft Excel and PowerPoint required

• Good oral and written communication skills a must
